Okay, not in any particular order :

Scan Disk
Defrag.
Virus check.
Trojan check.
Backup everything.
Run Adaware.

What else ? :-)

I do this on a Weekly basis. I save the once a month for bath, shave and clean
underwear change

Get rid of all the junk files.
MRU files
Temp files
and any others you can think of.

Uninstall programs you installed to test and never removed.
Clean up the registry.
Clean the computer. keyboard, mouse, mouse pad, monitor, the vents for
the air flow that cools the computer.
Adjust your chair to a comfortable yet practical position.
Check for latest security updates.
Go through your newsgroup ignore list. Add people. Remove people
Check for the latest version of the program you can't live without.
Eat :-)
Pay your electric and ISP bills
Got o work to get the money to pay electric and ISP bills :-)

Back everything up... don't forget your favorites/bookmarks like I did
once.

There is probably some more stuff as well.

I looked at that, and it seemed to pretty much be the canonical list.
Thus my first reply, about changing your sheets. :)

But then it hit me (as I was reading someone else' reply to your
question) that other than backing up maybe every couple of months, I
don't do any of those things. No, seriously -- it really did make me
blink. We get so used to how we work, we don't really *notice* our
routines (or at least don't really *think* about them) -- or the lack of
them.
 
Update virus definitions and ditto for AdAware.

everything else a minimum of weekly, sometimes every few days.

Man, do I feel clean all of the sudden! (Except for the sheet
thing....guess I'll just burn these and get some that don't stand at
attention).

Looks fine, could add:

Use your computer for computing !

You might want to "consider", before doing the above,

-Remove applications you'll never use
-Look for updates of most favourite programs
-Check for browser helper objects (Ad-Aware cover this?)
-Check cookies
-Check registry for errors, manually fix ones
that you understand, caution with the rest,
(use Powertools and OleClean).
-Take a 10 minute break at least, have some tea.
-Check view/options/filetypes for "dead wood", open regedit
and manually get rid of unwanted context entries. (right click)
- Manual backup registry, compact registry (caution)
- Empty temporary folder, recycle bin, cache, etc
- Run "sequoiaview" - examine folders taking up too much space
- Organise Start menu, bookmarks etc...
- Organise personal documents and files etc...
- Take a 10 minute break at least, have some tea.
- Go to the toilet.
- Look over start up group/s
- ...do the Scandisk/Defrag/etc you mentioned
- Read a magazine on the latest hardware you wish you had.
- Run System file checker, get new file CRC's (\windows\*)
- Get yourself some nice background wallpaper for your desktop
- Quickly e-mail a friend or write a thankyou to a software author
- Run "wizmo" and send your VDU into power saving mode
- Take a 10 minute break at least, have some tea.
- Partition, install a bootloader and start installing Linux !
- Sweat profusely...
- Go to bed tired out from nervous exhaustion, twitching.
