One can never have too many tweak utilities. ;-)

Yes, you can, though. One thing I expereinced was that they
sometimes they mess things up because they all tweak with
altering similar registry entries. There are many
"coreferences" within the registry. Some of them, even to
tweak the same thing, will use slight different part of the
reg. (e.g., the user vs. all user); conflict can happen and
things can be messed up.

So IMO, to avoid conflict, I would just use one tweaker. And
it is good to do some backup if you use them frequently.
