Probably not. That's one of the reasons for
Beta releases - to find out what works and what
doesn't.
 
You won't know unless you try or tell us which software you are referring
to. Most applications should work just fine, but some might require patches,
updates or new versions. Compatibility for 64-bit version is the same, your
applications need to be fully 32 or 64 bit native, no 16 bit applications
allowed.

If you are a model train hobbist remember there is still a lot of very old
16bit software being used, it will not and can not be made to work with Vista
now or ever, so it is nenessary to keep an XP machine.
 
Will Vista run 16bit software?

Some say,

Vista 32bit version will run:
16 bit software
32 bit software

Vista 64bit version will run:
32 bit software
64 bit software

Often driver, or software patches may be needed.

Is the right or wrong?
 
I need to do a little testing and get back to you about 16 bit software
support on Vista x86, as for 32 bit, there is no sure guarantee, since some
software might break because of changes to security in the system, so some
apps might not function well under UAC or operate with limited priviledges
or protected mode in IE 7+, things to take into consideration.

I have a copy of Works 4.5, Hallmark Card Studio 1.0 and Publisher 95, will
give them a try on Vista x86 and update you.

Vista x64, like XP Pro x64, will not run 16-bit software.

A lot of older 32-bit software used a 16-bit version of installer software
(the vendors never got around to updating their old reliable installers).
This is a frequently seen cause of incompatibility with x64 systems. In
other words, such programs could run but can't be installed.

I think it has to do with the Windows on Windows 64 emulation layer in the
x64 systems that allows 32-bit software to run. This of course does not
exist in Vista x86.

The answer seems to be that there is no simple answer yet, early days still.

16bit Model Train Control Systems:
All model train folks using 16bit systems, it maybe best to just buy a
modern second hand PC capable of running Vista Beta2, try 512MB video memory,
with 4GB RAM and run Vista Beta 2 and see if the software will install and
run.

Do NOT try Vista Beta2 on any of your production machines, not even as a
duel boot system. Treat Vista Beta2 as a hobby on a spare machine.

16bit and x86 32bit (old) Office and Business applications:
There really should not be any public need to run old office software
suites, just buy the lasest Vista ready systems, Office 2007, etc.
