So what exactly are my options for profiling my Kodak 8500 dye-sub?

I didn't like the initial result - big brightness jump from 245 to 255
(screen is profiled using the Spyder), and colour slightly off (but not
by much).

Tried tweaking 'Transfer' in the driver, but it has NO effect
('override' ticked). Still cant understand why.

Colour adjustment panel is a bit crude - brightness, contrast, gamma and
colour sliders only.

So, what is the best DIY procedure for creating a profile? Do I edit a
copy of an existing profile (8500 Matte or 8500 Glossy)? How do I do
that?

I've ended up with a couple of adjustment layers that I drag onto each
image in Photoshop before sending it to the printer. Works, but hardly
neat!

I'm happy to do a bit of 'trial and error' and look at test patterns
printed, but just don't know how to start.

Any pointers appreciated!

You could try editing the current profiles. The only thing I have found that
allows this, that is also free, is LABWARP from
http://www.eddiem.com/photo/labwarp/labwarp.html

It may not be able to edit your profile at all, but it is free so worth a
try.

Next you could try something like Monaco EZ color to create a profile from
scratch. Not exactly the cheapest method, but I have had decent luck with it.
After that look for hardware based solutions from companies like Xrite.

As far as reading material, I'm looking for the same things. Most of what I
had is gone after a fire. So I have to find some good references to put on my
shelf. Most likely I'll end up at the book store this weekend.
