No. The slide layouts are fixed and can't be changed. However, if you set
up a slide just the way you want, you can duplicate and edit the content and
the layout will match. You can do this for all new slides.
You can edit the MASTER SLIDE(s) to modify the layout to meet your needs. Go
to VIEW >> MASTER >> SLIDE MASTER. Note: you cannot add additional,
editable, text boxes to the Master Slide (limited to the title box and 1
content box).
If using PPT 2002-2003 you can create multiple Master Slides with various
layouts and mix-and-match as needed in your presentation.
