When using the FrontPage 2002 Web Search component, is it possible to search
all of a domain except for a particular folder?

Yes/No.

The search component can only search within the web it is inserted into, it
will not search across subwebs or search content in a folder that has a name
starting with the underscore character.

If you are hosted on IIS and have access to Index Server, then you can
create or the host can create a index catalog that can includes/excludes
content within the domain.
