Is there a way I can write a value to the registry that will prohibit/allow
a user to open a file based on the value of the key?

Would this change for 2000/XP and Me/98?
 
I dont quite get it.
How would I be able to say prevent a user from opening
"C:\test.txt" with a value placed in the registry?

I guess you have to move it to an NTFS partition and set the access rights.

You can't. As Armin said, set the NTFS permissions in the ACL of the
file when it's created.
