Normally no unless it is stamped or identified as reconditioned.

Otherwise you would need a new card to compare the card in question by using
a high power magnifying glass and look for either sloppy workmanship or some
areas that may have used a different soldering technique or discoloration of
the PC board near some component when comparing to the new card. Even this
may not reveal anything as I just went through this process a few months
back and could only suspect (not certain) that the cooling fan was replaced.

It should have a suffix on the part number, but
reconditioned can mean many things. A card that is sold and
returned in an unopened box is still used and may be sold as
is, as used or as reconditioned. A card that had been used
might be washed and tested or it might have chips or
fans/heat sinks replaced.

Suppose some of the components on the board have a black coating on them.

The edge connector pads will be unmarked if the card is brand new.
Whether the card was "reconditioned" or just "reboxed" would be less
obvious as "reconditioned" may have many levels.
 
Compare it to a new one.
