I have 30 users on a good lan using AD profiles. They have both a DC and the
server holding the profilesshare on the same lan. Sometimes when the users
logs in the profiles will not be loaded, but most they works. How can I
search this problem?

Usually when a profile is unable to load you will receive a popup error
stating why. Do you receive any errors? If so what is the error? You
should also check the application log for errors and post them in the group.
You can also enable userenv logging to obtain more details. See the
following article on how to enable it.

Detailed information - It dos not work copy a file that already is there.

After that message a new message shows that the user profile can't be
loaded, you are loging in locally.

This comes only sometimes for different users and different machines.
Sometimes it works for them.
