Is it possible to lock an amount (such us 10%) of the whole available
space on a flash drive (pen drive) to eliminate any possibility of
overwriting/cancellation/.... ?
How do i perform that?

Not clear what you mean there, you'll be told if you try to write more
than will fit, it wont just mindlessly overwrite what is already on it.

No. Unless you want to prevent 'soft' overwriting (within the filesystem)
only.

How do i perform that?

Then you can just partition the stick and make the partition smaller
than the whole space.
