Could somebody tell me at what point users are effected by setting the
pasword complexity requirement in Group Policy - i.e. in a live network if I
enable this requirement will users become locked out? Will they be asked to
change their passwords next ime they log on or will they only have to change
when the password period has expired?

Password complexity only takes effect when the users change their password,
either from password timeout or from the administrator setting the "User
must change password at next logon" option. So you can force them to use new
stronger password by setting that, or you can wait for them to change their
password and then they'll be required to use a stronger password.
