outlook enterprise (2007)

I run outlook enterprise (2007) under vista enterprise. I can get mail but
cannot send it. I have stopped defender and checked all the settings,
contacted my ISP . The settings seem to be correct, there is no port
conflict. Still can't send any mail.
Very frustrations; help please.
