Is there a way to restrict users access to directories on their local
computer using group policy? For example to deny access to %systemdrive% and
%systemroot% or maybe deny access to everything except their profile.
 
Yes, using the file system computer policy.

\Computer Settings\ Windows Settings\ Security Settings\ File System

(I think that's the path - I'm not in front of a server :-)
