When I click the Check of Updates button, it sayd it found no new definitions
or engine updates.

If you go to Windows Update, are you offered signature updates for Windows
Defender?

Are you on a corporate network?

I am not offered signature updates, but when I click Check for Updates, I get
the feedback mentioned in the original post.

This computer is on a corporate network. I have not encountered any other
errors.

If you are on a corporate network, access to Windows Update may be
restricted by policy. In addition, AutoUpdate, which is the mechanism used
within Windows Defender (i.e. when you go to Help, about, and press Check
for Updates)--may be routed to a corporate server.

There are several possible mechanisms that may be used--SUS, WSUS, or 3rd
party solutions. Of these, Windows Defender signature updates are
distributed via WSUS. However--this is a new class of updates, available
since January 16th, and the WSUS administrator must enable them.

Let me be sure that I understand what you are saying:

Were you stating that you are able to reach Windows Update (start, all
programs, Windows Update (or Microsoft Update)?)

And that when you do reach Windows Update, you are not offered signature
updates for Windows Defender, even though it is installed?

And that when you check for updates within Windows Defender, you get the
feedback that no updates are available?

I believe the last is expected if you are on a corporate network which
distributes patches via an internal server, and has not yet authorized these
updates--or perhaps doesn't have the correct infrastructure to do that.

However--the Windows Update behavior I don't understand. You should either
be able to reach Windows Update and have signature updates offered, or not
reach Windows Update, as I understand it.

When I check for updates within Windows Defender, I am told no updates are
available.

I haven't tried Windows update, but will. I just did, but had to install
the latest windows genuine advantage validation tool, so will reboot and try.
