Is the line count in MS word based on a 65 character line?

No. The line count would depend on the font, the actual text written in that
font, and the distance between the margins.
Even with fixed space fonts, there is the font size and the distance between
the margins to consider .

You sound like a transcriptionist. Unfortunately, Word defines "line count"
as a physical line count instead of a character-per-line count. To get a
usable (billable) line count, you have to use the character count and divide,
unless you can find a support app online that will do the math for you. Good
