How to remove mpeg file from system

Downloaded a mpeg video file from a popular meta video site. Try to delete
this file from system but encountered this message:
cannot delete <filename>: it is being used by another person or program.
Close any program that might be using the file and try agin.

Could it be a trojan or spyware running in the background?

I've encountered this on one or two occasions myself - it looked scary and
sinister, but in fact always turned out to be Windows getting itself into
some kind of loop that I never understood. In every case the problem
disappeared, but sometimes took more than one system restart. (Don't ask me
for an explanation!)

I never tried Engel's safe mode suggestion, but I would, if it happened
again, and a restart didn't do the trick.
