How can a letter in a heading in,e,g, in Windows Explorer be turned into a
hotkey?

If you are referring to the menu headings, are you not seeing the
underlined letters? If no, try pressing ALT on the keyboard. They
should then show up. Then press the underlined letter to extend the
menu or to perform that action.
