Check your temp folder assignment:
1. Right click on 'My Computer' and choose 'Properties'.
2. Click 'Advanced' and then 'Environment Variables'.
3. Change the temp variable to %USERPROFILE%\Local Settings\Temp.
NOTE: This actually showed to be correct on my system.
However, after pressing the OK button in the 'Environment Variables'-dialog
(instead of cancelling out) the value gets resaved.
This corrected my Error 101 issue and MS Anti-Spyware is now running fine!
