I am looking into setting up a server for remote secure file access that
will also act as a domain controller and Microsoft Exchange server. Is it
advisable to have these on the same machine, or will this cause problems?

Ewan/Captain Blammo-

It is not advisable to set up Exchange on a domain controller. This goes
against MS recommendations and general Best Practices. Also... you
generally want to avoid giving users access to your domain controller
(especially remote users).

My advice: You need 4 servers. 2 domain controllers. 1 Exchange server.
1 VPN server.

Agreed, Exchange and VPN servers should not be run on domain controllers.

Best not to have Exchange on a DC if you can avoid it, but I've done it in a
lot of small company networks.

Don't use this server for RRAS/VPN, tho - if you want VPN, my vote is with
something like a Sonicwall firewall to handle this.

Yeah, right, don't install Exchange on a domain controller.......
unless you're are using Small Business Server and have no other choice!
