Should DNS and AD be running on more than one DC in one domain?

Yes. You should have DNS on each DC.

No, one DC per domain is sufficient. Also, one single DNS server in your
whole infrastructure will work. However, there are many reasons to add more
DC's and DNS servers. For example, second DC and/or DNS provide the
redundancy if the first one fails.
