In my office I want an employee to be able to create a file, read, modify,
but to save it only on the hard drive. Is it possible to disable floppy and
USB ports so that only the Admin could enable them?

Disable them in the BIOS and password protect the BIOS.

You could disable them in the BIOS of the computer.
