Deleting Write-Protected Program

I am trying to delet a virus infected program, but when I try to delete it, I encounter the following message,
"Cannot delete sys; access is denied. Make sure the disk is not full or write-protected and taht the file is not currently in use." It is Window/System32/shdoclc.dll/dnserror.htm. It controls the Internet Explorer, and link automatically to "Searchmyrequest.com."Can any one tell me how to delete this file?

http://www.safer-networking.org/ download Spybot Search and Destroy from
there and do a full system scan.
