No, but it may make the defrag faster if you do.

Defrag has to back up whenever any running program writes to the
disk. In Safe Mode fewer programs are runnng than in normal mode.
Therefore defrag will usually finish faster in Safe Mode.

In Safe mode since only the basic processes are loaded, chances of
something interrupting the defragmenter in its job are less, hence its
a good option, although some third party tools will run in the
background even while the system functions.
 
when I defrag I do not run any programme. I set the computer for defrag and
wait till the job is completed.
in that case I presume it is not necesary to go into safe mode fro
defragmenting.

Many programs are running. A program that you initiate is just the
tip of the iceberg.

Hit Ctrl-Alt-Del and look at the Processes tab.
