If you would like to roll your own schedules with regard to scanning, use
the Windows Scheduled Tasks facility, or the AT command.
in \program files\windows defender, you'll find
mpcmdrun.exe:

Scan - Runs System Scan
-ScanType config, 1 (quick) or 2 (full), default 2
-Privileges restricted or full, default full
SignatureUpdate - Runs Signature Update

So--you can initiate quick or full scans, preceded by a signature update, if
desired, at any time you wish. I've just run a signatureupdate, and the
command apparently doesn't return until the check is accomplished--the
machine was, in fact, up to date, but it took a good long time--minutes--to
complete.
