Click on Start>Run and type DISKMGMT.MSC and then within disk-manager, you
can re-assign drive letters by right clicking the drive in question and
selecting "Change drive letters and paths..."

Yes. If you have installed some software through the respective drives and
then next time you run them, the software might look for data/CD in that
specific path and obviusly since it is changed and that path does not exist,
there may be problems. It is best recommended to leave the drive letters as
it is if they are not causing any problems to you, other than cosmetic
problems.
