With the 5400 first gen.:

In Minolta Scan Utility:

Preferences:

* Auto-Exposure for slides: ticked

* Output: 16 bit linear

* Focus: manual, with the knob:

With the film emulsion down in the holder, start the focus process with
the knob at the left extreme. Turn slowly to the right (holder moves to
the right). You will encounter a first peak. Go past that a bit. The
bars will fall back a bit, then do a second peak. That second peak is
the optimum focus. My observation/speculation is that the first peak
occurs when the scanner focusses on the back of the film, and second
when it focusses on the emulsion itself.

* Ice (and Grain Dissolver) on

Steps:

1. Prescan

2. Focus on light textured grainy area, near point of interest, and a
little away from centre of the slide, to compensate for slide
curvature.

3. In Exposure Control tab, move master slider down to -.1 or -.2 (MSU
auto-exposure tends to blow out highlight detail a bit too much for my
taste)

4. Scan

In Vuescan, scan-from-disk from the 16 bit linear generated above. Use
"Image" settings. Use a very slight, or zero clip of white and black
point. Color balance can be improved by using Vuescan's scanner profile
function to produce and use an ICC profile, with a target slides such
as those offered by Wolfe Fauste. I find "neutral" color balance works
well, in conjunction with this profile.

Or in Photoshop, assign same file Minolta's "posi linear" profile, then
convert to Adobe RGB, and work with levels/curves to your taste.
