For some reason, only one of my two speakers is producing sound. They're
plugged with a single cable into a single speaker connection on my desktop.
Is there some setting to be changed or other obvious explanation?

Several things to do. Before turning on the computer, check
the wiring to the speakers. Make sure that the plugs are
inserted properly and seated firmly. Also check on the
connection from the computer to the speakers for the same
thing.

With the computer on, open the mixer panel and look at the
balance settings for the sound output and the input. They
should, at least, be centered.

To check if the silent speaker is defective, switch them
around, i.e., left connected to right plug and vice versa.
Still silent in the "silent" speaker?

Results will determine the next course of action.
