It looks like PCI 2.2 allows for 3.3V cards as well as 5.0v cards. If
it is a 3.3V card, it will be keyed differently and will not physically
fit into your PCI 2.1 slot. Or, at least that's how I read this:

Since the enhancement listed for PCI 2.2 relies on that 3.3V service, I
would speculate that if the card fits in your slot, it will work.

Perhaps I misundertand what "keyed" means in the context of these cards.
I thought that refers to the posts and notches in the slot and cards
that orients the connection, but it might just be what pins are live. I
must admit that I've never had a PCI card not fit into the slot, and
I've put a lot of new cards in old machines.
So, I guess my revised answer is: Maybe?
